In cooperation with the compound management group from Boehringer Ingelheim (Biberach, Germany), a novel well geometry was developed to enable perfect positioning for even small sample volumes and facilitate the direct transfer from storage to assay plate.
An optimal well design to directly address small sample volumes was determined in practical trials. A flat well bottom allows precise pipette tip positioning with aspiration close to the well bottom. With a standard microplate height (ANSI/SBS 2-2004) and 0.5 mm sealing rim, common plate handling processes such as heat sealing, piercing, stacking and transportation are easily employed without laborious robotic adaptation. A well geometry combination consisting of a square-top, conically tapered to a flat-bottom, renders the ideal design to utilize a high working volume with minimal waste of sample material.

Inducible heat shock protein 70 (Hsp70, or Hsp72) is a stress protein whose expression is upregulated when the cell or organism is placed under conditions of stress. Hsp70 is essential for cellular recovery, survival and maintenance of normal cellular function. As a molecular chaperone, Hsp70 prevents protein aggregation and refolds damaged proteins in response to cellular stress caused by environmental insults, pathogens and disease. Hsp70 is also released into the circulation from both necrotic and non-necrotic cells, and its levels in serum are increased by a number of stressors. Hsp70 is believed to serve as an immunological "danger signal," and measurement of Hsp70 in serum and plasma is a useful tool in the study of heat stress, infection, cardiovascular disease, exercise, and psychological stress.

PEROXsay™ is a colorimetric quantitative peroxide assay that measures the oxidation of ferrous (Fe2+) ions to ferric (Fe3+) ions. Peroxides react with a sugar alcohol converting it to a peroxyl radical that subsequently starts the oxidation of ferrous ions to ferric ions. The acidic pH of the PEROXsay™ assay allows the ferric (Fe3+) ion to complex with the colorimetric reagent, resulting in a change in absorbance that is proportional to the peroxide concentration. The assay has a Linear Range: 0-50µM
PEROXsay™ is suitable for the following applications; quantifying the level of protein damaging peroxides in biological solutions, measurement of lipid peroxidation of low density lipoproteins and liposomes and monitoring protein glycation.

The recent discovery and characterization
of small non–protein–coding regulatory RNAs,
such as microRNAs (miRNAs), PIWI–associated RNAs (piRNAs
and rasiRNAs), short interfering RNAs (siRNAs), trans–acting
siRNAs (ta–siRNAs), and other families of short RNAs
has led to a rapid expansion of research directed at elucidating
their expression patterns and regulatory functions(1,2).
Currently, Northern blotting is the standard method for
the detection of small RNAs, because it allows direct comparison
of the quantity of small RNA between different samples.
However, major drawbacks of Northern blotting are time consuming
procedures and poor sensitivity, especially when monitoring
expression of short nucleotide sequences. Despite the
improvements in detection sensitivity provided by Locked
Nucleic Acid (LNA) substituted probes(3), Northern blotting
requires relatively large amounts of starting material and
involves multiple handling steps (Table 1).
Another approach for small
RNA detection is based on the ribonuclease protection assay
(RPA), which takes advantage of liquid hybridization kinetics
to improve detection sensitivity (Table 1). Both Northern
blotting and RPA determine the amount of small RNA by measuring
the amount of the labeled probes that are non–covalently
hybridized to the small RNA, providing a relative quantification
of small RNA levels. In addition, the critical experimental
points that determine the assay efficiency, which are the
hybridization and wash steps in the Northern Blot assay
and the hybridization and ribonuclease treatment in RPA,
usually require optimization because insufficient
treatment may lead to non-specific detection while excess
treatment may compromise the detection sensitivity.
USB offers a novel approach for direct labeling and detection of small RNAs. The miRtect-IT™ technique takes advantage of liquid hybridization kinetics to improve detection sensitivity, but without a procedure that requires optimization. Similar to Northern blotting, miRtect-IT™ does not require specialized equipment or any amplification step, and thus allows direct and accurate measurement of specific small RNAs.
