Laminin is an extracellular matrix protein which contains a number of functional domains that allow it to assemble into sheets. Trevigen's mouse laminin is purified from EHS sarcoma.
The PathClear® designation means that in addition to standard sterility, endotoxin and MAP testing, the Laminin is tested by PCR and is clear of 31 pathogens and viruses, including LDEV. Each lot is rigorously qualified in biological performance assays.

MACHEREY-NAGEL, a leader in purification kit innovation and manufacture, has answered the need for reliable RNA purification out of various starting materials.
NucleoSpin® RNA II is suited for fast and easy extraction of highly pure RNA out of cultured cells and various tissues like e.g. paraffin embedded tissue, biological fluids, bacterial cells (Gram-negative and Gram-positive).
High yields up to 70 µg ready-to-use RNA can be obtained.
RNA purified with NucleoSpin® RNA II is suitable for applications like real-time PCR, RT-PCR, Northern blotting, primer extension, array technology and RNase protection assays.
Filters and quantitative DNA removal for best possible results
NucleoSpin® Filters (shredders) are part of the kit and cause an efficient homogenization and reduction of viscosity.
Using the rDNase included in NucleoSpin®
RNA II the resulting RNA is without detectable genomic DNA.
In fig. 1 gel-electrophoresis results of RNA purified from five
million HeLa cells are shown. On a denaturating formaldehyde
gel (1A) the RNA shows a band pattern typical for HeLa cells,
consisting of 18S rRNA, 28S rRNA and a nuclear precursor RNA
(n.p. RNA) which is sometimes misleadingly interpreted as genomic
DNA. The high molecular weight band indicates highest RNA quality
as precursor RNA is very prone to degradation.
Fig. 1b shows the same RNA sample in a TAE gel which stains
DNA considerably stronger than denaturating formaldehyde gels.
The RNA fraction has been treated with RNAse in order to get
a clear view on possible DNA contamination in the RNA elution
fraction, which however cannot be detected.

Fig. 1: a) On the denaturating formaldehyde agarose gel the RNA eluate shows not only the 28S and 18S rRNA but as well the nuclear precursor RNA (n.p. RNA). b) Using non denaturating conditions and having digested the RNA only the degraded RNA can be detected but no chromosomal DNA.
Accurate highly sensitive results
NucleoSpin® RNA II allows the purification of highly pure RNA suitable for accurate results in demanding downstream application, like RT-PCR.

Fig. 2: Agarose gel of RT-PCR products using
different amounts of HeLa cells
In Fig. 2 a RT-PCR of total RNA isolated with the NucleoSpin® RNA II standard protocol is shown starting with 105, 104, 103, 330, 100, 33,10, 3 and 0 HeLa cells. The RT-PCR was done using 3 µl of total RNA each (elution volume 40 µl) and ß-actin primers. The specific RT-PCR product is 626 bp of size. The results show that the recovery from the NucleoSpin® RNA II is as efficient that RT-PCR is possible with only RNA from 10 HeLa cells.
Outperforming the competitors kits
Fig. 3 shows purified total RNA from 106 HeLa cells in comparison to competitor Q. PCR reactions were performed for 30 cycles using different sensitive gene-specific primers. An aliquot of the PCR reaction was analyzed using a 1% TAE agarose gel.

Fig. 3: Agarose gel of PCR products after RNA
extraction using the NucleoSpin® RNA II (MN) and a competitor
product (Q) for the detection of chromosomal DNA contamination
Even when small targets are amplified no DNA contamination can be detected using the MACHEREY-NAGEL kit whereas the kit from competitor Q shows obvious contamination resulting in false positives.
NucleoSpin® RNA II provides you best results in your sensitive downstream applications.
NucleoSpin® RNA II offers a full solution
including the DNase and filters for optimal results.
