Associates of Cape Cod Inc. – 1→3-ß-D-Glucan: Pharmaceutical Contaminant and Biological Response Modifier

American Pharmaceutical Review™ article re-publication.

The establishment of consistent safety and efficacy of parenteral pharmaceutical products is a key goal in GMP operations.  Within the spectrum of activities that support GMP operations, the control of bioactive contaminants is a critical concern.  Practically, and perhaps principally, this has meant sterility and within-specification endotoxin burdens.  Over the last seven decades, tests for bacterial endotoxin burdens have developed from exclusively animal-based bio-assays to LAL (Limulus Amebocyte Lysate), or LAL-derivative, biochemical assays and mammalian cell-based assays.  These methods have been enshrined in national and international regulatory requirements and have performed extremely well in maintaining a high level of endotoxin-related safety in modern parenterals.  This success has not engendered complacency, however, and pharmaceutical science relating to bioactive contaminants has moved forward to identify other bioactive materials whose uncontrolled presence in parenterals can potentiate risk to safety and efficacy.  One such commonly encountered bioactive is 1→3-β-D-Glucan.

EpiCypher – Anatomy of a Recombinant Nucleosome

Nucleosome substrates are emerging tools for the in vitro study of chromatin and epigenetic regulation, with applications from basic research to high-throughput drug screening.  The advent of recombinant designer nucleosomes (dNucs) carrying fully defined covalent modifications of the histone proteins (e.g. lysine acylation) or wrapping DNA (e.g. 5’  methylcytosine) enables novel or dramatically improved approaches (e.g. reader binding, enzymatic assays, antibody profiling) for next-generation chromatin research.  This document proposes the minimal thresholds for the assembly and quality validation of these reagents; essential if they are to yield reproducible, biologically-relevant data.

Hycult Biotech - Neutrophil Elastase in Ocular Graft-Versus-Host Disease

The purpose of this study was to compare neutrophil elastase levels in tears of oGvHD patients with normal subjects.  14 oGvHD patients and 14 matched normal controls were enrolled in the study. Neutrophil elastase (Product HYBHK319) was measured from tear washes of both eyes.  The results demonstrated that NE-levels of oGvHD patients were 250-fold higher in comparison to the matched controls.  Secondary markers such as MPO, MMP-8 and MMP-9 were also significantly elevated in the oGvHD group.  This study shows that NE can be a potential marker of ocular inflammation.  However, the effectiveness of NE as an inflammatory biomarker remain unknown.  Large, well-designed prospective studies are needed to test the effectiveness of an NE assay.
